Laboratory application of the dextran-coated-charcoal radioimmunoassay of insulin.

چکیده

A modification of the method for radioimmunoassayof insulin is described, which achievessatisfactory sensitivity while employing commercially available reagents and antiserums. These modifications consist mainly in lowering the initial percentage binding, reducing the amount of labeled insulin, and preincubating the unlabeled hormone with antiserum. Application of the linear isotope dilution formula, modified for our conditions, proves to be a reliable substitute for the standard curve.

Modification of a commercially available digoxin radioimmunoassay kit for higher precision.

The procedure prescribed for the Schwarz/Mann 125-l-digoxin kit was modified with regard to pipetting and counting procedure. Incubation time with dextran-coated charcoal was also investigated. The modifications resulted in a significantly higher precision wihout introduction of a systematic error and the analysis time per eight-tube batch was decreased by 20 min.
